Rosmarinic acid: a potent carbonic anhydrase isoenzymes inhibitor
نویسندگان
چکیده
Rosmarinic acid is a water-soluble ester of caffeic acid and 3,4-dihydroxyphenyllactic acids, and is mainly found in plant species including Boraginaceae and Lamiaceae. In this research, we determined the inhibition property of rosmarinic acid on carbonic anhydrase isoenzymes I and II (hCA I and II) purified from human erythrocytes by using Sepharose-4B affinity column chromatography. hCA I and II isoenzymes were obtained with a yield of 57.9% and 67.2% and 76.5and 509.3-fold purification of each isoenzyme, respectively. In order to show the purity of the isoenzymes, SDS-PAGE was performed and one band was observed. In vitro inhibition of both hCA I and II isoenzymes by rosmarinic acid using CO2 -esterase activity gave Ki values of 86.0 μM and 57.0 μM, respectively.

Isolation of Human Carbonic Anhydrase B and C and Apocarbonic Anhydrase by Affinity Chromatography
Human carbonic anhydrase B and C have been purified in high yields by affinity chromatography, using two different affinity gels, incorporating the potent benzenesulfonamide inhibitor, but coupled to Sepharose through different ,spacer-arms~. Although both affinity gels bind carbonic anhydrase B and C quantitatively, they exhibit different binding capacities. Sepharose-glycyi-L-tyrosine-azobenz...
متن کاملCapsaicin: a potent inhibitor of carbonic anhydrase isoenzymes.
Carbonic anhydrase (CA, EC 4.2.1.1) is a zinc containing metalloenzyme that catalyzes the rapid and reversible conversion of carbon dioxide (CO2) and water (H2O) into a proton (H+) and bicarbonate (HCO3-) ion. On the other hand, capsaicin is the main component in hot chili peppers and is used extensively used in spices, food additives and drugs; it is responsible for their spicy flavor and pung...

The isoenzymes carbonic anhydrases B and C (EC 4.2.1 .l. C02tH20 2 HCO; t H+) from human erythrocytes have been extensively investigated by different physico-chemical methods in order to understand the mechanism of action of these efficient enzymes [l-4]. These two enzymes differ in their catalytic rate by a factor of about 5 for a number of reactions investigated. The primary structures of the...

Co(II); Zn(II) and Cu(II) complexes of two new sulfonamide carbonic anhydrase (CA) inhibitors, derivatives of thienothiopyran-2-sulfonamide, were prepared and characterized by analytic, spectroscopic, magnetic and conductimetric measurements. The new complexes are more potent CA inhibitors than the parent sulfonamides, with IC(50) values around 0.1 nM, against isozyme CA II.
متن کاملStudy of Glycation Process of Human Carbonic Anhydrase II and Investigation of Effect of Fasting On Enzyme Activity by Using Spectroscopic Methods
Background: Glycation is the non-enzymatic reaction between the carbonyl groups in sugar and free amino groups in proteins. this reaction leads to changes in structure and functions of proteins. Advanced glycation end products (AGEs) is the final stage in this process, which is highly oxidizing and destructive nature, causing many diabetic complications. Methods: In the present investigation, ...
